Getting true tone from your guitar speaker may require breaking in your guitar speaker or Guitar Speaker Break-In. While there are different recipes for doing this, it is not recommended to pull it out of the box and play a loud gig.

Getting the best tone out of your speaker and getting it to match the tone of its tone samples usually takes time.  You only have one chance to break a speaker in.  If you miss that chance when it is brand new, it may never have the intended tone, or your desired tone that you create by your playing style.  The video below explains how not all guitarist agree with this.

Celestion

Celestion has a “how to” that you can follow at their website.  Google “how to break in a guitar speaker” – and their article comes up first.   Since Celestion is the flagship guitar speaker manufacturer, it is no surprise that this seems to me to be the best article that I have found on this topic.   It was written by Dr. Decibel himself, their main technical contact.

Jensen

The word from one sound engineer and speaker manufacturer is that Jensen’s newly manufactured vintage speakers require comparatively little break-in time.  So consider this when implementing the above instructions.

Weber

Jensen’s competitor in that niche – Weber speakers – are known to have a very long break-in time which may require more complicated instructions.  You will need to find out about that before purchasing one.

Others

Check the manufacturer website regarding guitar speaker break-in requirements and/or recommendations before purchasing.  They may be more than you have time for – or want to bother with.
